細胞培養を用いた胃粘腫防御機構に関する研究

研究概要

1、ラットおよびラビットの単層培養は寺野、太田らの方法(Gastroenterology83:1280ー91,1982)によた。
2、 ^<51>Cr放出試験による細胞のviabilityの測定は寺野らの方法(Gut25:19ー25,1984)に従った。
3、プロスタグランディン、cAMPなどの測定はRIA法によって行った。さらにHPLCにより産生されたPを行い、PGE2,PG12などが主に産生されていることを認めた。
4、培養細胞の粘液分泌および産生の測定は ^3Hーglucosaminなどの取込をみた。
5、Free radicals発生モデルおよびそれによる細胞障害ならびにその消去剤による影響の研究を行っradical産生系として、xanthineーxanthine oxidaseの系を用い、あるいはエタノ-ルによるfree radi生モデルを作成し検討した。この結果free radicalのうちhydroxy radicalが細胞障害にもっとも重要有し、その消去に粘液が重要な役割を有していることが認められた。
6、内因性PGの産生の制御機構を、細胞内情報伝達機構の観点から検討した。この結果、PGの産生nd messengerとして、Ca^<++>イオンが重要な役割を果たしていることが判明した。cAMPの系はPG産生にていなかった。
