夏型過敏性肺臓炎の発症機構に関する免疫学的研究

研究概要

夏型過敏性肺臓炎の発症機構を免疫学的に解明する目的で、本症の原因抗原であるトリコスポロン・クタネウムの発症抗原をモノクロ-ナル抗体を用いて分離精製することを試みた。まず、発症抗原に対するモノクロ-ナル抗体を作成するために、本真菌の環境分離株であるTIMM1357株(serotype I)とTIMM1318株(serotype II)の培養濾液抗原をそれぞれイオン交換クロマトグラフィ-法およびゲル濾過法を用いて、分子量100万前後の高分子多糖体分画を得た。この高分子多糖体分画が菌血清型関連多糖体であること,ならびに発症抗原を含んでいることを夏型過敏性肺臓炎患者における吸入誘発試験および家兎を用いた実験モデルで確認したのち、この抗原をBALBICマウスに免疫して脾細胞を得た。次に、この免疫して得た脾細胞をPAー1ミエロ-マ細胞と細胞隔合を行い、この細胞から数種類のモノクロ-ナル抗体を得た。Serotype Iに特異的に反応するモノクロ-ナル抗体はIgG1に属し、Serotype IIに特異的に反応するものはIgG2aに属し、両Serotypeに共通して反応するものはIgMに属していた。現在、これらのモノクロ-ナル抗体を活性型担体であるLargeA LargeFーTresylーTOYOPEARL 650Mに結合させたカラムを作成し、このカラムに菌血清型関連高分子多糖体をアプライして、発症抗原を精製中である。今後、このようにして得られた精製抗原を用いてヒトならびに実験モデルを用いて本症における免疫学的発症機構を検討する予定である。
