IgEリセプタ-を介する肥満細胞の増殖機構に関する研究

研究概要

肥満細胞は細胞膜上にIgEリセプタ-(FceR)を多数有しており、FceRをIgE分子あるいはanti-IgEで架橋することにより脱顆粒が起こることが知られている。われわれは、従来終末細胞と考えられていた、マウス腹腔中の成熟組織肥満細胞がin vitroで活発に増殖していることを見いだした。この増殖にはインタ-ロイキン3(IL3)IL4の両者の持続的な存在が必要であり、IL3とIL4の作用機構も今回の検討で明らかとなった。さらに組織肥満細胞の増殖に対する、IL4の作用はFceRを抗原特異的なモノクロ-ナルIgE分子とそれに対応する2価以上の抗原で架橋することにより代用しうることが明らかとなった。このFceRの架橋によるシグナルはIgE分子とanti-IgEによる架橋によっても同様に発揮された。2価以上の抗原を用いて起こる肥満細胞の増殖は1価の抗原を培養系に加えることにより濃度依存性に抑制されることからFceRの架橋によるシグナルが組織肥満細胞の増殖には必要であると考えられた。FceRの架橋により肥満細胞からIL4などの因子がautcrineされ、これが肥満細胞の増殖に関与している可能性も考え、マウス腹腔細胞よりPercoll比重遠沈法を用いて成熟肥満細胞を分離し、これにIgE分子と2価以上の抗原を添加し培養し、終時的細胞を回収し、Northern blot hybridizationによりIL4mRNAを検索したが今回の検討では検出できなかった。また、IL4による組織肥満細胞の増殖のシグナルはプロテインキナ-ゼの活性ガングリオシド物質であるTPAによっても代用しうることが明らかとなった。IL4とFceRの架橋、TPAはそれぞれ細胞内シグナル伝達機構が異なることが明らかになったが、3者はいずれも肥満細胞の増殖に働くことから、今後、さらに詳細な検討が必要と思われる。
