ヒト血液細胞におけるIgEレセプタ-発現調節におよぼすサイトカインの役割

研究概要	低親和性IgE受容器(FcεRII/CD23)およびその放出体である可溶性FcεRII/CD23はIgE産生調節に関与したり、IgEの関与する免疫応答での機能的受容器として働くと考えられている。本研究はこのCD23の発現調節にインタ-ロイキン4(IL-4)やインタ-フェロン(IFN)が深く関与していること明らかにした。すなわち、ヒト単球細胞U937やヒト好酸球細胞株EoL-3において、IL-4やIFN-γはmRNAレベルでCD23の発現を増加させた。IFN-γはBリンパ球においてはIL-4によるCD23発現増強を抑制し、IFNの作用は細胞の種類により異なった。IFN-αもCD23発現を増加させたが、CD23mRNAの増加は少なかった。逆に、副腎皮質ステロイドはこのmRNAの発現を強く抑制してCD23発現を減少させた。Transforming growth factor βもCD23発現を抑制したが、mRNAレベルでの抑制は弱かった。このように、本研究はCD23の発現調節機構を明らかにし、CD23発現を調節してIgEの関与する免疫応答を人為的に調節する新しい治療法を展望するものである。一方、血清中の可溶性CD23はその大部分がBリンパ球より放出されると考えられ、年少児ほど高値を示し、加齢と共に次第に減少した。アトピ-性疾患を有する乳幼児は非アトピ-対照児に比較して有意に高い血清値を示したが、年長児ではこの両群間の有意差は無く、血清可溶性CD23値は乳幼児でのアトピ-疾患のマ-カ-となりうることが明かとなった。さらに、生後にアトピ-疾患を発症した児の、帯血可溶性CD23値は発症しなかった児に比して有意に高く、帯血可溶性CD23値により生後のアトピ-疾患の発症予測が可能であると考えられた。
