核抗原の細胞表面への発現とその病因的意義

研究概要

本年度の上記課題「核抗原の細胞表面への発現とその病因的意義」について以下のような研究結果を得ることが出来たので、既提出の実施計画に沿い報告する。
1)新生児包皮由来の表面細胞の無血清培養を、従来用いられていた牛下垂体抽出物を用いずに、安定的に継代培養することが出来た。
2)成人皮膚から得た人工表皮を用いる培養は、MCDB153培地のみでは安定的継代培養が出来ず、牛下垂体抽出物の添加により3継代までの培養が可能となった。
3)新生児包皮由来の培養細胞を用いて、紫外線(主にUVB)による影響を調べたところ、細胞表面への抗核抗体特に抗SS-A/RO、抗SS-B/La、抗RNP抗体が特異的に結合することが判った。この現象は、Flow cytometryにより定量的に観察出来、線量依存性で、glycosylationに依存し、細胞周期とは無関係であることが判った。
4)更に、同様に抗RNP抗体の結合を、エストラダイオ-ルで刺激した培養細胞で検索したところ、有意の結合性の上昇がみられた。興味深いことには、抗SW抗体はほとんど結合性を示さなかった。
5)次に、プロスタグランディンJ_2で培養細胞を刺激すると抗RNP抗体の細胞表面への結合が誘導されることが判った。ヒ-トショックプロテインあるいはストレス蛋白の誘導と抗ENA抗体の細胞表面への結合性の関係を知る上で、きわめて重要な事実と考えられた。
6)ヒトリンパ球とENA発現培養細胞との混合培養を行ったが、現在のところ、細胞生障害涯性は観察出来得ていない。
今後、同様な研究を培養した血管内皮細胞を用いて行う予定である。これらの研究により、ヒトル-プスエリテマト-デスの紅斑の発症の機序の一部が明らかになるのではないかと思われる。
