天疱瘡における水疱発生機序の分子生物学的研究

研究概要	1.ウロキナ-ゼのインヒビタ-であるPlasminogen activater inhibitorー1(PAIー1)による天疱瘡新生児マウスモデルにおける水疱形成阻止の実験を行なうため,培養ヒト表皮ケラチノサイトの培養上清よりPAIー1の精製を行なった。無血清培養法の系では、培養ヒト表皮ケラチノサイトは48時間で約50ng/mlのPAIー1を産生するが、精製には不十分なため培養ヒト表皮ケラチノサイトによるPAIー1の産生を増加させる条件を検討した所、TGFーβlの添加(5ng/ml)により、増殖は著明に抑制されたが、PAIー1の産生は約20倍の増加が認められた。TGFーβを添加した培養液2リットルより、ConーA column及びPAIー1に対するモノクロ-ナル抗体を用いたAffinity columnにて精製した所約500μgのPAIー1を得た。しかし、これは不活性型であるため、SDSによる活性化を試みたところSDSを除去するステップで阻止活性が10分の1程度に低下した。このため当初予定した、新生児マウスモデルの実験は行なえなかった。今後は、精製開始時の培養上清量を増加させる事および活性化にVitronectinを用いることを検討する必要があると考えた。 2.抗マウス・ウロキナ-ゼ抗体を作成する目的で、マウス表皮ケラチノサイトのCell LineであるPAM Cellの培養上清より、精製を試みた。PAM Cellの培養上清には48時間で約5IU/mlのウロキナ-ゼが分泌され、5リットルの培養上清よりBenzamidine Affinity Columnを用いて、Batch法にて精製を試みたところ、約5μgのマウス・ウロキナ-ゼを得た。現在、これを用いて家兎にて抗体を作成した。 3.新生児マウスの表皮より、Total RNAを分離し、マウス・ウロキナ-ゼDNAをテンプレ-トしたProbeを用いて、Northern blotによりマウス・ウロキナ-ゼmRNAの同定を行なったところ、新生児マウス一匹分の表皮より同定が可能であった。
