Elマウス発作発現機構における脳内グルタミン酸とγ-アミノ酪酸の役割に関する研究

研究概要

Elマウスは放り上げなどの体位変換刺激によりけいれんが誘発される自然発症てんかん動物である。われわれはElマウスの抑制性ならびに興奮性神経伝達物質の異常に関して研究をすすめてきたが、今年度はElマウス大脳皮質及び海馬切片におけるグルタミン酸の取り込み並びに放出について検討を行なった。
実験方法:実験動物はElマウスとその母系のddYマウスを使用した。Elマウスは生後3週齢より、常法に従って1週間に2回放り上げ刺激を行ない、毎回連続してけいれんをおこすEl(+)と、放り上げ刺激を行わないEl(-)について実験を行った。グルタミン酸の取り込みと放出はChapmanらの方法によった。神経伝達物質の放出は高濃度K(50mM)刺激による放出と定義した。
実験結果と考按:El(-)のグルタミン酸の放出は、大脳皮質においても海馬においてもいずれもddYより高値であり、一方El(+)における放出はEl(-)より低値であった。この際、El(-)とddYとの間の相異は遺伝的に規制されているものであり、El(+)とEl(-)との間の相異はEl(+)における反復するけいれんの結果に基づくものと想定される。Elマウスの海馬におけるグルタミン酸の取り込みは、El(+)もEl(-)もともにddYに比べて非常に低値であった。しかし大脳皮質においてはいずれのグル-プ間にも有意差が認められなかった。大脳皮質のグルタミン酸の放出は50mMK^+-灌流液に1mM及び5mMグルタミン酸を加えることにより促進するが、10mMのグルタミン酸を負荷するとグルタミン酸の放出は抑制された。また、海馬切片へのグルタミン酸の取り込みも、高濃度のグルタミン酸を負荷することによって低下した。これらの実験結果はグルタミン酸の放出と取り込みはオ-ト・レセプタ-系によって調節されていることを示唆するものである。
