肝糖新生系の新しい調節機構とその異常

研究概要	肝の糖新生系は,さまざまな肝外因子と肝内因子によって調節を受けており,血糖の調節に重要な役割を果している。本年度は糖新生系が,肝の脂肪酸代謝によって,どのように調節されているかを検討した。 (1)Sodiumー(+)ー2ー[6ー(4ーchlorophenoxy)hexyl]ーoxiraneー2ーcrrloxylate(Etomoxir)は,脂肪酸分解の律速酵系であるcarnitine palmifoyltransーferase Iの特異的阻害剤である。この物質を絶食ラットに投与すると,肝からのグルコ-ス放出が抑制され著しい低血糖症に陥った。 (2)1988年,金沢大学より低血糖症,脂肪肝を呈するマウスが報告された。この低血糖症がいかなる原因によっておこるのかを検討したところ,血清,肝,骨格筋,心臓の遊離型カルニチン,long acyl carnitine,short acyl carnitineの含量が低下していた。総カルニチン含量は,疾患モデルマウスの場合,対照に比し約20%であった。次にカルニチンを生理的食塩水に溶解し投与したところ,低血糖症は改善した。このような全身性カルニチン欠乏症のモデルマウスの報告はなく,脂肪酸代謝異常と肝の糖新生系の関係を分析するには,有用なモデルマウスと考えられた。
