卵胞の発育・分化に関与する新しい卵胞内因子の同定とその生理作用に関する基礎的研究

研究概要

我々は、これまでにブタ卵胞液中にインタ-ロイキンー2レセプタ-/p55(ILー2R/p55)誘導因子の存在を発見し、 1.本因子が卵胞液中に存在するだけでなく、ブタ卵巣顆粒卵巣顆粒膜細胞で産生される。 2.卵胞液中のILー2R/p55誘導活性は、および顆粒膜細胞からの産生が形態学的・機能的に卵胞の発育とともに低下する。 3.卵胞液中のテストステロン/エストラジオ-ル比とILー2R/p55誘導活性の間に有意な正の相関関係が認められ、本因子が卵胞の閉鎖に関連する可能性がある。 4.本因子は分子量25,000から43,000の蛋白性高分子である。 5.ヒト卵胞液中のILー2R/p55誘導活性は、抗インタ-ロイキンー1αまたはβ抗体によって中和されないことから、ILー2R/p55誘導活性を有するインタ-ロイキンー1(ILー1)ではなく、全く未知の因子であろう。 6.卵胞液中に高濃度に存在するエストラジオ-ル、プロゲステロン、テストステロン、アンドロステンジオンにはILー2R/p55誘導活性が認められないことを明らかにした。さらに、ILー2R/p55誘導活性の点で本因子と密接に関連するILー1の卵巣内局所調節因子としての機能についても検討を行い、ILー1がブタ顆粒膜細胞培養系において、増殖促進作用があり、LHによる形態適学的・機能的黄体化を抑制するだけでなく、今年度は新たに、未熟ブタ顆粒膜細胞に対するFSH作用に及ぼすILー1の影響について以下の知見を得た。
1.ILー1はFSHによるアロマタ-ゼ誘導およびLH誘導cAMP産生を濃度依存的に抑制する。
2.FSHによるLHレセプタ-誘導およびプロゲステロン産生を抑制する。
以上より、ILー2R/p55誘導因子およびILー1が卵胞の発育・分化に密接に関連することを証明し、これらの因子の卵巣局所調節因子としての可能性を示唆した。今後、これにの因子の生理的意味付けについて検討を続けたい。
