研究概要

エアロゾル粒子の分析・性状測定:石炭フライアッシュの粒径密度分離法を改良して密度1.8〜3.2g/m^3の各画分の化学組成を明らかにした。NiOの各メーカー試料についてESR,XPS特性を調べた。レーザー光散乱パルスとICP発光パルスから、粒子の粒径と元素組成を関連させるための装置を試作した。表面に不均一な帯電をもつ壁面へのエアロゾル粒子の沈着特性を実験的に検討した。鉛・食塩粒子の凝集体について画像解析・化学分析を行いその形状特性を検討した。
吸入粒子の呼吸器内挙動:肺胞を模擬した伸縮球内でのエアロゾル粒子の混合・沈着とエアロゾル流れの流体力学的特性との関連を実験的に明らかにした。イヌ・ラット・ハムスターの呼吸器内吸入粒子沈着量計算モデルを作成し、とくにイヌについてはより詳細な葉部計算モデルを作成して、沈着量の動物種間差、葉部間差について検討した。
吸入実験システムの開発と吸入実験:各種ニッケル化合物(NiS,Ni_2O_3,NiO,Ni)粒子によるラットの暴露実験を行い、各臓器中の滞溜率とその粒径依存性について検討し、化学形態と有意な関連があることを明らかにした。食塩粒子を核としてDOSを主剤とする単分散粒子(粒径0.7〜1.0μm)の吸入実験システムを作成し、ヒトによってその有用性を確かめた。粒子状物質(炭素粉または大気塵)と二酸化ちっ素ガス混合物によるラットの長期暴露実験(30ケ月)を行い、ガスと粒子との影響が相加的であるとの知見をえた。小動物に対する鼻部吸入実験装置と吸入中の動物の呼吸機能測定装置を完成し若干のデータをえた。また気道内挿法によって性状・粒径の異なる粒子をラットに投与し、気道内の部位別・粒径別粒子挙動についていくつかの知見をえた。
