腫瘍抗原の分子免疫学的研究

研究概要

免疫系によって認識されるがん抗原の性状および腫瘍化に関連する遺伝子を解析し、次の結果を得た。1.正常と同一の一次構造を持つGM3ガングリオシドであつてもある特定のGM3密度において、T細胞や抗体が認識できるメラノ-マ抗原性を獲得できることをGM3-リポソ-ムを人工抗原として用い証明し、がん抗原に新しい概念を導入した。GM3密度を上昇させる物質として24KdのGM3結合蛋白を同定した。2.蛋白性メラノ-マ抗原遺伝子をクロ-ン化し、塩素配列を決定したところ、内在性レトロウイルスのenv.遺伝子産物であることが判明した。この遺伝子発現は正常組織では全く見られず、腫瘍化に伴って発現するものと考えることができた。この遺伝子をL細胞に導入発現させることでメラノ-マ抗原性が出現するところから、この分子が抗原発現に重要な役割を担っていることが判明した。上記1.2.の結果を考え合わせる自己に内在する分子ががん抗原の活性化等に伴って持続的に発現し、それが免疫系によつて認識されるがん抗原分子として働くものと考えられた。3.腫瘍化、増殖に関連する遺伝子のcDNAクロ-ンを単離し、塩素配列を決定した。M末側にTFIII-A、Kruppel等に見られるZn finger構造を有し、実際DNAに結合する活性を有する全く新しい遺伝子であることが判明した。ノザンブロット解析ではほとんどの腫瘍細胞株においてこのmRNAは出現するが、正常組織では未熟精子母細胞、脳、副腎以外での発現は認められなかった。脳では胎児脳で最も強く発現し、成熟するにつれて発現が低下する。このことから、この遺伝子はDNA結合活性をもつ蛋白であり、転写制御/細胞増殖/組織発生過程に重要な働きを持つものと考えられた。
