酸化還元因子ADFの解析と新しい抗リンパ球活性化抗原抗体によるIL-2Rの解析

研究概要

IL-ZE/p55(Tac)の誘導因子として報告されたADF(ATL-derived facor)はcDNAクロ-ニングの結果、強い内因性還元物質であるthioredoxinのthomologueであることが明らかになった。組換えADFは、それ自身強い還元活性を持つ他、IL-2R/Tac誘導活性に加え、PBMC.B.T細胞株のヌイトジエン、IL-2反応性を高めるCo-cytoklne活性を持ち、細胞活性化の新しいメカニズムの存在を示唆すると考えられる。ADFのアミノ酸配列の31番目のCys基をSer基に置換した還元活性を持たないmutant ADFを作製した。現在、還元活性と各種生物活性の関係を検討中である。さらに、a)ヒト胎児および成人胸腺髄質、正常リンパ・節の間質細胞にADF高発現が認められた。ADFがリンパ系の発生分化過程においてなんらかの役割をはたしている可能性が考えられる。b)HTLV-I,EBV,HPVトランスフォ-ム細胞および子宮頸部HPV陽性癌化部位にADF発現が増強していた。c)中波長紫外線を反復照射すると正常ヒト表皮のほぼ全層および培養正常ヒト表皮細胞にADF発現が増強した。さらに、マウスにX線照射後ADFを反復投与するとADF非投与群に比べて延命効果が認められた。以上のようにADFは、(1)癌化の過程を組織化学的に解析する手段として、(2)細胞の活性化機構の新しいメカニズムを与える可能性を持つ多機能物質として、(3)種々のストレスに対する生体防御因子としての可能性を持つ物質として重要であり、現在ADFに関する基礎的な解析を進めている。2.単離LGL細胞に対して、YTA-1抗体が増殖誘導、キラ-活性の誘導作用を持つ。125I-anti-IL-2R抗体(TU27,Tac)を用いた実験よりYTA-1抗体がIL-2R/p75の選択的なinternalizationを促進する事が示された。また、YTA-1抗原はIL-2R/p75分子とは異なることが明らかとなっており、IL-2Rシステムのシグナル伝達の制御分子としてYTA-1抗原の解析が重要であると考えられる。現在我々はYTA-1分子のN末端部分アミノ酸配列を決定し、そのsequenceの確認とそれに基づくcDNAクロ-ニングを行いつつある。
