グルココルチコイド作用増強物質によるリンパ性白血病の新治療法の開発

研究概要	我々は、(1)標的細胞におけるグルココルチコイド(GC)ホルモンの感受性を増強する細胞感受性増強物質群、(2)標的細胞でのGC作用の発現を増幅する作用増幅物質群、(3)標的細胞でのGC作用を抑制する物質群を見出し、これらを用いてGC感受性リンパ性白血病、リンパ肉種症への応用を試みてきた。GC作用を増幅する生体内活性物質は、プロテインキナ-ゼCの活性化作用を持つジアシ-ルグリセリド(DG)であったが、本年度の研究で、スフィンゴリピットの代謝産物、スフィンゴシンがGC作用を著明に抑制することを見出した。しかもスフィンゴシンはプロテインキナ-ゼCの特異的阻害剤である。このように生体内ではフォスファチジ-ルイノシト-ルの代謝産物のDCと、スフィンゴリピッドの代謝産物のスフィンゴシンが細胞レベルでGC作用をそれぞれ増強、抑制していることが推定された。さらにプロテインキナ-ゼCの阻害剤のH-7やスフィンゴシンが初代肝細胞培養系、肝切片組織培養系でGCレセプタ-の核内移行を抑制し、Cytosol画分に熱ショック蛋白のHSP-90と結合した状態で留まることを明らかにした。以上の結果を基に、現在GC感受性リンパ性白血病の新しい機序に基づく治療法の開発を試みている。DGは、生理血中濃度のGC作用を著明に増強してL5178Yリンパ肉種の増殖を抑制するが、中でも炭素鎖18のDGが最も強い効果を示し、炭素鎖8,14のDGと有意の差を示した。しかしこれ等DGのプロテインキナ-ゼCの活性化能はほぼ等しい。またこれらのDGはいずれも下垂体からACTHの分泌量を変化させず、間脳下垂体-副腎間の調節系を障害しない。このように調節因子の脂肪酸の組成の違いによりこれ等の因子の細胞親和性が存在することが考えられ、副作用なしに標的細胞のみでのGC作用の増強、抑制が考えられ、これ等の可能性を現在検討している。
