研究概要

細胞複製に連動した、遺伝子発現の包括的制御を分子の水準で解明することを目標として、細胞生産装置なかでも転写装置と翻訳装置の形成と機能制御の解析を進めてきた。本年度は、下記の成果が得られた。
1.大腸菌染色体に存在するすべてのプロモ-タ-の同定、強度順位の決定とサイクリックAMPなどの転写因子に対する感変性を指標とした分類を進め、転写シグナルの全体像にさらに接近した。
2.転写装置の基幹成分RNAポリメラ-ゼ各サブユニットの分担機能、サブユニット蛋白上の機能ドメインを同定する研究を引続き進め、機能地図の概要を作成した。
3.熱ショックなどのストレスを加えた時のRNAポリメラ-ゼの機能変化の構造基盤を明らかにしたのに続いて、複合要因による増殖静止期への移行時には、転写装置・翻訳装置が貯蔵型に変換することを発見した。
4.RNAポリメラ-ゼの機能制御に転写因子との相互作用があることを先に実証したが、転写因子の活性調節に蛋白リン酸化が関与することを発見した。
5.翻訳装置リボゾ-ムの機能制御に関与する「リボゾ-ム修飾因子」(RMF)を発見した。また、リボゾ-ム蛋白の修飾に関与する遺伝子の生理機能の探索を続けた。酵母ミトコンドリア・リボゾ-ム蛋白の遺伝子12種を単離し構造を決定した。
6.マイコプラズマの全tRNA遺伝子を同定した。また、真核生物の天然サプレッサ-tRNAを発見し、翻訳制御の新しい機構を明らかにした。
