研究概要

1.研究の目的:光エネルギ-受容タンパク質の光化学系2複合体は約20種類のタンパク質成分と数種類のクロモファアで成分で構成されている。本年度の主要研究目的は、光化学系2反応中心複合体の主要構成成分であるD1タンパク質とD2タンパク質および光化学系2複合体においてMnクラスタ-を保持する役割を担っている表存性33ーkDaタンパク質の構造と機能発現の解明とすることである。
2.本年度の研究成果:(1)光化学系2反応中心複合体(D1/D2/Cytb559)をオクチルグリコシドで処理して、クロロフィルとフェオフィチンを結合しているD1/D2タンパク質複合体を単離し、その光化学的・物理化学的性質からこのヘテロダイマ-が反応中心を形成していることを明らかにした。(2)D1タンパク質の前駆体のC末端を切断してD1タンパク質に変換するプロセシング酵素を精製した。(3)D1タンパク質のMn結合部位を同定するために、エンドペプチタ-ゼで処理して得られるD1タンパク質断片をMn2+アフイニティクロマトグラフィにかけ、その結果からMnを結合するアミノ酸配列を推定した。(4)光化学系IIコア複合体を種々のエンドペプチタ-ゼで処理し、43ーkDaタンパク質と47ーkDaタンパク質を特異的に欠く系II複合体を調整した。これらの標品への33ーkDaタンパク質の結合の様相から、33ーkDaタンパク質の結合する内在性成分はD1/D2/Cytb559であることを推定した。また、これらの標品が酸素発生能を保持していることから、43ーkDaタンパク質と47ーkDaタンパク質は酸素発生反応には直接関与していないことが明らかとなった。(5)遊離状態の33ーkDaタンパク質と光化学系IIの内在性成分に結合した状態の33ーkDaタンパク質とをそれぞれ化学修飾し、その結果から33ーkDaタンパク質分子内の結合ドメインを推定した。
