研究概要	遺伝性運動失調症のモデルとしてミエリン形成不全症マウス(myelin deficient,mld)を選び、分子遺伝学的解析を進めた。mldではMBP遺伝子が重複しており、上流の遺伝子の一部が逆転している。本年度はMBP発現低下の原因、ならびにmld座の形成機構について検討した。その結果、以下のことが明らかとなった。 1.MBP発現低下の原因 1)mldでは上流のMBP遺伝子の逆転した部分が転写されることによりアンチセンスRNAが生成されること。 2)アンチセンスRNAは主に核内に存在し、センスRNAと相補鎖を形成すること。 3)上流の遺伝子から下流の遺伝子の転写開始部位に到達する転写産物はごく微量であること。以上のことから、下流の遺伝子の発現抑制のメカニズムとしてはアンチセンスRNAがセンスRNAのプロセッシングまたは細胞質への移行を妨げることが考えられた。 2.mld遺伝子座の形成機構 1)mld遺伝子座の約110kb、正常マウスMBP遺伝子を含む約85kbの領域がコスミドを用いてクロ-ニングされた。 2)mld遺伝子座内には3個のjunction pointが存在し、上流の遺伝子と下流の遺伝子との間には1.4kbの由来不明の繰り返し配列が挿入されていること。 3)正常遺伝子における各junction pointの相当部位の間で明らかな相同性は見られないこと。以上のことから、mld遺伝子座は重複、逆位、未知のスペ-サ-配列の挿入を伴う多重組換えの結果生じたものと考えられた。
