鳥類視蓋における位置特異的分子発現の決定

研究概要	二次元的にきっちりと対応のつく網膜-視蓋投射の形成過程で、視蓋の細胞に位置特異的分子が発現されて、それを視神経線維が確認するといわれている。本年度は、そのような位置特異的分子の発現の決定は孵卵2日目にすでになされているかどうかを、ウズラ視蓋原基を180°回転してニワシリ胚に移植することにより追究した。網膜の局所的ラベルにはカルボシアニン系色素DiIを用い、会載標本で検索した。視蓋原基を180°回転することにより得られたキメラ脳は手術がうまくいくと、奇形になることはないが、移植片よりできた(ウズラの)視蓋は反対側のホストの視蓋よりも常に小さく、組織の最終的な大きさは遺伝的に決まっていることを示している。正常では網膜鼻側からの線維は視蓋尾側に、網膜耳側からの線維は視蓋吻側に投射する。孵卵2日目で10体節期頃に視蓋を180°回転して、視神経線維の投射を見ると、正常と同様に網膜鼻側からの線維は移植された視蓋尾側に、網膜耳側からの線維は視蓋吻側に投射していた。移植片の同定には、蛍光観察の後パラフィン包理を行い、切片にFeulgen染色を施すことにより行った。本実験の結果は、視蓋の吻尾軸は10体節期頃まではまだ決定されておらず、視蓋原基を回転しても調節がおこることを示している。本実験では移植片が回転された状態を保っていることを確認するために移植の際Di0を用いてラベルした。実体顕微鏡オリンパスS2M-111を用いることにより移植の効率をあげることができた。また薬用保冷庫MPR-211Fは場所をとらずに効率よく標本を保存することができた。
