ショウジョウバエ分子生物学による生体高次機能解析

研究概要	以前の研究ではトランスポゼ-ス活性を欠くQ系統であるWY113を用いてPサイトタイプ形成に重要な役割を果すP因子を単離した。このP因子はORF3を欠失している為完全型P因子が体細胞に於て産生しているORF2までの蛋白とほとんど同じものを産生していると考えられた。これらの結果からリプレッサ-の本体はこのORF0,1,2からなる蛋白であると結論づけた。本年度は以下の研究結果を得た。(1)第3染色体上に存在するこのP因子を抽出した系統を作制し、そのリプレッサ-活性をGDsterilityおよびsn^w二次突然変異率で測定したところ、これらの系統は一年後でももとの系統ほど強いPサイトタイプを形成しなかった。その為、強いサイトタイプの形成にはORF2までのP因子ばかりでなく他の弱い活性を持つP因子の働きも必要なことが明らかとなった。(2)ORF2までの蛋白単独で強いPサイトタイプを形成し得るかどうかを調べる為に、ヒ-トショックプロモ-タ-の支配下にあるORF3を欠失したP因子〔W^+,hsp△ORF3〕を作製し、いくつかの形質転換体を得た。これらの系統はsn^wの雌性不妊を誘発した。また、熱処理をしたものとしないものではサイトタイプ形成に明らかな違いが見られた。しかし、熱処理をしたものでも強いPサイトタイプは増さなかった。このことは、このP因子が確かにヒ-トショックプロモ-タ-の支配下にあり何らかの転移抑制因子を産生しているが、強いサイトタイプの形成には他の要因も必要だということを示している。(3)ヒ-トショックプロモ-タ-の支配下にある完全型P因子(I carus-neo)はPサイトタイプ中でもsn^wの二次突然変異を誘発したが、通常ヘルパ-PWCではry系統の形質転換を全くおこすことができなかった。これらの結果は、リプレッサ-は転写を抑制することでP因子の転移を抑制しているらしいということを示唆している。
