研究概要

今年度における本研究班の研究実績は次の通りである。
1.X、Y精子の分離については、無担体電気泳動法によってヒト精子をX、Yの2つのピークに分け、それぞれよりDNAを抽出し、アメロゲニン遺伝子のPCR法による増幅による判定を行った。
2.精子形成に関しては、培養ゼノパス精細胞におけるプロタミン合成と遺伝子発現を検討し、sp4遺伝子のcDNAをプローブとして、この遺伝子は第一精母細胞に発現し、精細胞になって翻訳されることをみた。
3.クローン化したニワトリLHBサブユニットのcDNAを用い、ウズラにおけるこの遺伝子の発現を環境条件との関連で検討した。またウズラ卵巣の受容体とLHの結合の基本的性質を明らかにした。
4.ラットの性腺刺激ホルモン分泌に応じた精巣セルトリ細胞の分泌能および細胞間コミュニケーションを、セルトリ細胞を培養して調べた。また精巣造精細胞の培養条件の検討を行った。
5.性腺発育障害要因を解析するため、キメラマウスを用いて実験を行った。C3Hマウス特異モノクロナル抗体を用い、C3HとBALB間で性腺の形態形成およびキメリズムを組織レベルで解析した。
6.性腺内免疫細胞の役割に関し、黄体内マクロファージがTGFーbetaを産出することにより、PRLによる黄体プロゲステロン分泌維持能を仲介していること等をin vitroの実験で明らかにした。
7.生殖系列におけるY染色体の役割を明らかにするために、長腕の反復配列を選択的に除外してシングルコピーDNAをクローン化するベクターの開発に成功し、Yの特定部位判定に用いる11種のプローブを得た。
8.コイ卵より卵成熟促進因子を部分精製し、分裂酵母のcdc2遺伝子産物とホモログなことを確かめた。その中の1つは卵成熟誘起ホルモン処理後に出現するので、成熟促進と密接に関連するものと思われる。
