研究概要

今年度の研究計画に従って班員の仕事は順調に展開している。その結果各班員がこれまで明らかにし得た事は(I)抗ZPーOポリクローナル抗体で観察されたと同様の受精阻止作用が今回確立した抗ZPーOモノクローナル抗体でもみられること(及川)、(II)卵管内では問題なく進行する胚発生が試験管内でうまく進行しないいわゆるin vitro 2ーcellブロックが低酸素下培養、Superoxide Dismutase添加により解除されること(森崇)、(III)テラトカルシノーマからDBAアガロースを用いたアフィニティークロマトにより糖タンパク質群を単離し、初期胚性の細胞に発現される新しい膜蛋白をいくつか同定したこと(村松)、(IV)in vitro系でマウスのMPにおけるロイコトリエンC_4(LTC_4)の産生能を、MPの形態、等を検討した結果マウスMPは従来報告されているよりも強いLTC_4産生能を有すること(舘)、(V)TTKー1由来の蛋白性免疫抑制物質の単離を試みGel上単一の物質までの精製に成功していること(森庸)、(VI)クラスI遺伝子L^dとK^bからプロモーターを取り除きニワトリ細胞質βーアクチンプロモーターに置き換えたもの(AcーL^dとAcーK^b)を作製し、マイクロインジェクション法によりC57BL/B6,C3Hマウスの受精卵に導入し、3匹のマウスにAcーL^d遺伝子が2匹にAcーK^b遺伝子が入っている事が確かめられたこと(宮崎)、(VIII)ヒト羊膜上皮細胞(FL細胞)を用いてフィブロネクチンへの接着をみると、レセプターを介した接着が成立すること(伊勢村)、(VII)カドヘリン分子の結合特異性決定領域の検索結果はP型カドヘリンのN未端113アミノ酸領域に特異性を決める部域が存在すること(竹市)、(IX)In vitro培養系における顔面形成異常に及ぼす影響をみると、尾体節数10個を境としてそれより遅い時期に卵黄のう開放を行うと全ての胎仔は正常発生することなどがそれぞれ明らかになった。
