研究概要

組換えワクチニアの系を利用し、HIVのgag遺伝子の単独発現でHIV様粒子が形成されることを発見した。またトランスジェニックマウスで、HIV遺伝子の発現がリンパ系組織、脳および網膜とレンズで強いことを見い出した(渋田)。HIVのrev遺伝子とHTLVー1のrex遺伝子ば共に非スプライス型のgagおよびenvのmRNAの発現に必要であるが、その塩基配列に相同性はない。しかしrexの標的配列のrevは作用しないがrevの標的配列にはrexも作用することを見い出し、revとrexの作用機作が極めて類似している事が予想された(吉田)。HIVのvif遺伝子はHIVー1とHIVー2の両者でウイルス増殖を促進する効果を持つが、vpr遺伝子はHIVー1においてのみウイルス増殖を正に制御していることを明らかにした。またHIVー1のtatとrevはHIVー2やサルエイズウイルス(SIV)にも作用するが、HIVー2やSIVのこれらの遺締子がHIVー1には作用しにくいことを明らかにした(足立)。HIVのリセプターCD_4の構造を反映すると考えられる抗イデオタイプ抗体を作製したが、それは外国で報告されたものと異なり、HIV中和活性を持っていた。今後の研究に有用であり、その大量精製法を確立した(大野)。HIVのgag蛋白質と逆転写酵素を大腸菌で大量に生産させ、それを完全に精製したが、これらは診断用の抗原として極めて特異性が高く、かつ研究用資材としても非常に有用である(中田)。HIVとSIVの各種の分離株の塩基配列の比較から分子系統樹を更に精密なものとし、HIVの起源と進化を考察している。またHIVー1とHIVー2のグループごとにenv蛋白質で保存されいてる領域を明らかにしワクチン開発の資料とした(五條堀)。以上のごとく、全ての班員のところで、着実で世界的にみても独創的な研究の発展が認められた。
