研究概要

本研究はコドン選択を決める諸要因を総合的に解明し、それらを基に遺伝子工学の基礎知識をも得ようとしている。原核生物遺伝子のコドン選択については、AT/GC圧のコドン選択への効果を知る目的で、ゲノムGC含量の極端に偏る生物種の遺伝子構造を解析した。武藤はリボソ-ム遺伝子を中心に、小笠原はDNA合成に関係した遺伝子を中心に解析を続けており、AT/GC圧の一般性を実証しつつある。真木は大腸菌mutT変異株を解析し、MutTタンパク質がAGミスペア形成を抑制する分子機構を解明して、AT/GC圧の実体の一つを明らかにした。武藤は池村と共同で、マイコプラズマのtRNAの定量を行い、tRNA量がコドン選択パタ-ンと密接に関係することをも示した。三田はカイコのユビキテン遺伝子の構造を決定し、このデ-タを基礎に、広範囲な真核生物のコドン選択にAT/GC圧が関与することを示した。池村は高等脊椎動物染色体DNAの巨大GC含量モザイク構造の存在を示し、そのモザイク構造がコドン選択パタ-ンを決める要因であることを解明した。和田と陶山は共同して、DNAの2重ラセンの物理的安定性が遺伝子ごとに一様化される傾向に着目し、この傾向のコドン選択への効果を明らかにした。陶山はこの効果とタンパク質生産量との関係についても解析を行っている。和田は2連字出現頻度のコドン選択への効果に関する知見をも得ている。熊谷は宿主細胞として放線菌を用い、同義コドンを入れ換えた人工遺伝子を作成し、コドン選択パタ-ンがタンパク質生産量に影響を及ぼすことを示した。放線菌のコドン使用頻度に基づき、人工遺伝子の大量発現系をも確立している。五條堀はレトロウイルスと肝炎ウイスル遺伝子のコドン選択パタ-ンと塩基置換速度との関係を解析し、複製酵素の性質と塩基置換速度の関係を明らかにした。
