研究概要

アルツハイマー病脳に特異的に出現する老人斑の重要な構成成分の一つであるアミロイド前駆体タンパク(以下APPと省略)の沈着するメカニズムを明らかにするために、まずマウスおよびヒトAPP遺伝子の構造が詳細に解析され、特に転写調節領域についてその発現制御に関与すると考えられる部位が明らかにされた。さらにこのAPPcDNAを培養細胞に遺伝子導入し、APPタンパクの代謝過程を詳細に検討できる実験系が開発された。老人斑を構成する1成分であるα1ーantitrypsinのヒトcDNAクローニングがなされた。アルツハイマー病の実験動物モデルの一つとなり得る老化促進モデルにおいて、血清高密度タンパクのアポタンパクの一つであるapoA-II遺伝子にアミノ酸置換を伴う点突然変異が見出され、アミロイド生成機構において重要な役割を演じていることが発見された。アルツハイマー病脳における遺伝子発現の異常を明らかにすることを目的として、ヒト剖検脳から良質のメッセンジャーRNAの抽出方法が確立され、さらにアルツハイマー病脳のcDNAライブラリーが作製された。家族性アルツハイマー病の遺伝子同定のための連鎖解析への戦略が検討された。また、in situハイブリダイゼーション法による遺伝子座の同定方法が報告された。家族性アルツハイマー病、ダウン症において重要な役割を担っていると考えられる第21染色体の構造を明らかにするために、第21染色体に含まれるNot I制限酵素認識部位の大部分をクローン化し、マッピングのためのマーカーとして極めて有用性の高いDNAクローンが得られた。さらに、染色体ソーディングにより第21染色体を単離し、目的のDNAマーカーを含む巨大DNA断片からクローン化する方法が開発された。さらにこの第21染色体からコードされる新たなタンパクが発見され、それが脳において発現していることも示され、その生理学的役割に興味が持たれている。
