研究概要

シナプスおけるインパルス伝達のメカニズムを明らかにする上で、新しい実験系の確立をはじめとしていくつかの課題を設定した。本年度は準備期間という位置付ではあるが以下に述べるような成果が得られた。(1)新しい実験系の確立:トランスミッタ-の放出を最も観測し易い実験系を確立することを目指した。その結果(イ)カメ錐体細胞、(ロ)シビレエイ電気器官巨大シンプトゾ-ム、(ハ)NG108ー15細胞(ニ)肥満細胞の4つに決定した。(2)膜融合観察システムの確立:神経終末部からトランスミッタ-が放出される場合、シンプス小胞と生体膜との融合が先ず起きる。その時小胞は消滅し、伝達物質はATPと共に細胞外に放出される。小胞の消滅は光散乱法で、また伝達物質の放出された場所と時間は、ルシフェリンールシフェラ-ゼ反応を利用して観察可能である。我々は映像信号同期方式を用いた顕微測光システムを用いて上記の実験を開始したがソフトの部分に不備の点があることが分かりその改良に相当の時間を費やした。しかしマルチショットパルスレ-ザ-顕微鏡は稼働しはじめた。更に2種の異なった画像を全く同時に見ることの出来るWー顕微鏡の分解能を上昇させることが出来た。レ-ザ-共焦点顕微鏡で細胞内Caを観察する方式もうまく作動するようになった。またこれを電顕レベルで観察出来るよう準備が完了した。
(3)この他の主な研究成果としては、(イ)ロイコトリエン、PAFによる神経機能の修飾機構の一部が判明した、(ロ)小脳プルキンエ細胞に新しいCaチャンであるPチャネルが見いだされた。(ハ)N型Caチャネル抗体の精製が開始された。(ニ)新しいAキナ-ゼ阻害剤とCキナ-ゼ阻害剤が開発された。(ホ)NMDAレセプタ-の特性が次第に明らかとなった。(ヘ)水銀面に展開することによりタンパク質の2次元結晶を作成する手法が確立した。
