多層的生体評価系による細胞成長因子利用手段の開発

研究概要

近年多くの成長因子・サイトカインが発見され、それらを新薬として実用化することへの期待が高まっている。しかしながら、これら新生理活性物質の生理作用評価系は細胞株を用いた評価系が主流であるため、その生体への有効性の検討が困難を極めており、成長因子・サイトカインの生体評価系の確立が急がれている。本研究では、多方面の研究分野において、生理学・病理学などのin vivo実験系を開発してきた一線の研究者により、成長因子・サイトカインの生体評価系の開発を行った。その結果・1)神経成長因子がテンカン発症を遅延させる効果を有すること、2)腫瘍成長因子-αが性腺刺激ホルモン放出ホルモンの分泌を制御する視床下部の電気活動を抑制すること、3)体外受精・初期胚操作の上で、大きな問題であった、培養系における初期胚発生停止現象が、アクチビンにより解除されること、4)アクチビンが下垂体の卵胞刺激ホルモン分泌細胞の分化増殖を促進すること、5)形質転換成長因子-βが妊娠維持に必須な卵巣からプロゲステロン分泌を促進する作用を有すること、6)チモシン関連ペプチドであるオビオペプチドが、原虫・細菌感染症の予防・治療に有効であること、7)ヒドラの刺胞反応を利用して、成長因子・サイトカインの超高感度測定系の開発に成功したことなど、成長因子・サイトカインが新治療薬として有効に作用する疾病についての新しい知見を数多く得る一方、新生理活性物質の評価系として極めて有効な生体評価系の開発にも成功した。
