パッチクランプ法にて記録される神経細胞電気信号自動解析用ソフトウェアの開発

研究概要	HITACコンピュ-タ-システムの演算処理速度を高速化するための電子回路を試作し、コンピュ-タ-のハ-ドウェアに追加して装備した。またこの処理に付属して高速演算用のソフトの開発を行った。これらの神経細胞電気信号自動解析用ソフトウェアを用いて、ラット副腎髄質クロマフィン細胞におけるムスカリン性およびニコチン性興奮に関係したイオン機構を解明するためのパッチクランプ法による研究を効率よく実施することができた。すなわち、アセチルコリンは単一チャネル電流を誘発し、この電流の振幅は膜電位依存性であり、逆転電位0mVであることを見出した。このアセチルコリンによって活性化される単一チャネル電流はヘキサメトニウムによって遮断されたがアトロピンでは抑制されなかった。ピペットに高K溶液を入れ、膜電位を静止膜電位レベルでクランプした場合には、短時間の開口をもった群発性単一チャネル電流をパッチクランプ法で記録した。チャネル開口の振幅および頻度は膜電位および外液のKイオン濃度に依存した。また、インサイドアウト法で記録した高Kによる単一チャネル電流はKイオンチャネル遮断薬であるテトラエチルアンモニウムを膜の内側に与えることによって抑制された。細胞アタッチパッチクランプ法においてムスカリンはチャネル開口頻度を用量依存性に減少した。これらの成績は副腎髄質細胞におけるムスカリン性興奮が静止膜電位レベルで開口するカリウムチャネルの抑制によっておこることを示すものであり、細胞電気信号自動解析における高速演算用ソフトウェアの開発によって、今回のパッチクランプ法を用いた研究を円滑に実施することができた。
