研究概要

昨年度は当該研究プロジェクトの基礎研究としてアフリカツメガエル卵母細胞の発現系の開発と初代培養肝細胞の新しい応答系の確立を行った結果、非肝実質細胞が肝細胞の増殖制御に重要な役割を果たしていることが判明した。そこで本年度は、ヒト単球(マクロファ-ジ)が産生する種々の因子群の検討を生物工学的方法で行った。
1.活性化単球に特異的なcDNAライブラリ-の作成。
ヒト末梢血から単球を大量に分離精製し、エンドトキシンであるリポポリサッカライド(LPS)で刺激培養した。LPS無刺激ヒト単球から調製したpoly(A)^+mRNAより、LPS(-)cDNAライブラリ-を作成し、LPS(+)cDNAライブラリ-間で差引きハイブリダイゼ-ションを行い、LPS刺激で発現される特異的なcDNAライブラリ-を得た。
2.In vitroでのmRNAの合成。
上記で作製した差引きcDNAライブラリ-からT3RNAポリメラ-ゼによりin vitroでmRNAを合成した。合成したmRNAの検定はインタ-ロイキンー1及びー6をプロ-ブとしたノザンブロット法で行い、それらが単球から分離したmRNAのサイズとほぼ同じであったことから合成されたmRNAが完全鎖であることが判明した。
3.マイクロインジェクションによる生物活性の検討。
合成したmRNAをアフリカツメガエル卵母細胞に注入し、培養上清の生物活性を検討した。インタ-ロイキンーの検定をMH60株細胞の増殖促進活性で調べると、添加した血清は用量依存的に細胞増殖を促進した。このことはバイオアッセイ系が正常に作動していることを示している。そこで、この培養上清を初代培養肝細胞系に添加してその機能変化を解析した結果、肝細胞に対する著名な生存促進活性がみられた。また上清の酸性処理でDNA合成の抑制活性が観察された。現在、これらの生理作用を示す遺伝子を探索中である。
