研究概要

私共はrasp21類似低分子量G蛋白質やその活性調節蛋白質を精製するとともにそのcDNAをクロ-ニングし、これらの蛋白質の機能や種々の病態との関係について解析を進めている。これらの低分子量G蛋白質にはGDP結合型の不活性型とGTP結合型の活性型が存在し、不活性型と活性型の転換を制御する蛋白質が存在すると考えられている。私共は低分子量G蛋白質の活性制御蛋白質として、smgp21とrhoAp21に対するGTPase活性促進蛋白質(GAP),smgp25Aとrhop21に対するGDP解離促進蛋白質(GDS)を見出した。さらに、smgp25AGDI,rhoGDI,smgp21GDSのcDNAを単離してそれらの一次構造を決定するとともに、その特異抗体を作製した。最近rasp21は、翻訳後、C末端のCysーAーAーXのCys残基にファルネシル基が結合し、次いでAーAーX部分が除かれ、さらにCys残基がメチル化を受けることが知られている。私共はC末端の構造がCysーAーAーXであるsmgp21BとrhoAp21の翻訳後修飾はrasp21と基本的には同じであるが、Cys残基にはファルネシル基とは異なるゲラニルゲラニル基が結合していることを明らかにした。また、C末端の構造がCysーXーCysであるsmgp25Aでは、少なくとも一方のCys残基にゲラニルゲラニル基が結合していることも明らかにした。これらの翻訳後修飾は低分子量G蛋白質の細胞膜への結合や活性調節蛋白質との結合に必要であった。また、種々の組織中のファルネシルトランスフェラ-ゼやゲラニルゲラニルトランスフェラ-ゼを測定するアッセイ法を開発した。一方、低分子量G蛋白質そのものについては、smgp21が血球細胞の分化やある種の白血病の成因と係わり合っていることを示した。また、smgp25Aが神経細胞の分化マ-カ-あるいは神経由来の腫瘍マ-カ-として有用である可能性も示した。以上の結果を得たことにより、本研究の研究計画はほぼ達成できたと考えている。
