脳機能改善薬を志向したリ-ド化合物の探索研究

研究概要

1.β-カルボリン類の合成:芳香性β-カルボリンのマンザミンCおよびユ-ディストミンH.I.Pの合成をトリプタミンとアザシクロウンデセンおよびプロリンより完成した。オキサチアゼピン環をもつユ-ディストミンL.F.CをN-ヒドロキシトリプタミンとシスティナ-ルより合成する方法を確立した。3,4-ジヒドロ-β-カルボリンのNbを活性化することで1-位のアルキル化が可能であることを見出した。2.スフィンゴ脂質などの合成:山崎グル-プが以前単離構造決定したセレブロシドB_1をニトロアルド-ル反応を利用して合成した。海洋産セラミドN-ラウロイルドコサスフィンガジエニンの光学活性体を合成し立体化学を明確にした。これら合成中間体を渡辺グル-プに提供できた。3.向中枢神経作用成分の探索:モノアミン酸化酵素B型に対してin vitroで10^<-7>〜10^<>-8Mで阻害作用を示した子のう菌より単離されたアントラキノン系のMSA、アザフィロン化合物TL-1はマウス静脈内投与では作用を示さないことが明らかになった。これらの誘導体を用いて作用を検討中である。一方グルタミン酸受容体への^3-Gluの結合に対する効果を指標とした活性成分の検定法の検討を行なった。マウス中脳などのP_2画分を用いて子のう菌、生薬などの抽出エキスの検定を行なうと混在する金属イオンの影響が大であることが明らかとなり改良法を検討中である。この他50種の子のう菌、30種の植物、生薬エキスを調製した。4.脳機能を反映する中枢性胃液分泌を指標にした脳機能作用:(1)数種の脳機能改善薬が胃液分泌を顕著に刺激することを見出した。ことにホパンテン酸塩、ニチコリン、TRHに強い作用が見られた。(2)グルタミン酸受容体刺激薬のカイニン酸に強い胃酸分泌刺激作用を見出した。(3)三環性うつ病薬の胃酸分泌抑制作用を見出した。(4)GABA受容体サブタイプとしてB受容体の胃酸分泌刺激機能を示唆し得た。麻酔薬も胃酸分泌刺激作用があることを見出した。
