DNAシ-ケンサ-による塩基配列決定の高速化

研究概要

本年度は計画年度の2年目であり、かつ最終年度であることから、昨年度までに開発を進めてきたミニトランスポゾンを用いての高速塩基配列決定法の総仕上げを目指し、開発してきた方法を実際の整列クロ-ンに適用して「実地テスト」を行ない、方法の評価やさらに改良すべき点の検討を行なった。一方、この方法を、大腸菌ばかりでなく、パン酵母のゲノム解析に適用するための基礎となる研究も進めた。
本年度の研究の結果、(1)ミニトランスポゾンを効率よく大腸菌の整列クロ-ンに導入するとともに、容易に独立な導入体を得る方法を確立し、(2)それぞれのミニトランスポゾン導入体に含まれる大腸菌の染色体を、ミニトランスポゾンおよびラムダファ-ジベクタ-の右腕あるいは左腕との間でPCRを用いて増幅し、得られた増幅断片をクロ-ンすることなく、塩基配列決定に用いるための方法を確立した。さらに、(3)このようにして開発した方法をパン酵母のゲノム解析にも適用するために、パン酵母ゲノムの約15%を網羅する整列クロ-ンバンクを確立し、それとともに、(4)上述したミニトランスポゾンを大腸菌およびパン酵母の両者で選択可能とするために、パン酵母の遺伝子であるLUE2をミニトランスポゾンに結合したものを作成し、(5)これを用いてパン酵母での組織的な遺伝子解析を行ない、同時に高速で塩基配列決定を行なうための方法の確立と大腸菌株の作成を行なった。一方、(6)DNAシ-ケンサ-を用いた塩基配列決定それ自体についても、デ-タ解析ソフトの大幅な改良を図り、それによって塩基配列決定の精度を落とすことなく、解析速度を少なくとも5倍程度向上させる方法を確定することができた。
