慢性疼痛並びにモルヒネ耐性における可塑的神経回路変調と可視化

研究概要

一つは、神経因性疼痛における可塑的機能変調の分子機構の解明とその可塑的変調の可視化を試みてきた。
神経因性疼痛モデル動物における神経因性疼痛の責任線維を分子レベルでの解析から同定した。この線維の特異的発現受容体のプロモーターによって制御される蛍光蛋白質遺伝子を脊髄後根神経節に特異的に発現させることに成功した。この手法により、責任線維の可塑的シナプス形成を可視化することを試みており、結果がで次第、論文作成に取りかかる予定である。また、責任線維の脊髄後根神経細胞のみをターゲットにした神経傷害時の可塑的遺伝子変調の解析も試みている。一方で、神経因性疼痛における可塑的機能変調の分子機構は神経傷害後に誘導されるリゾフォスファチジン酸が一次知覚神経の脱随、後根神経節や脊髄後角における痛み関連性遺伝子・蛋白質の発現調節を行うためであることを突き止め、論文投稿中である。
もう一つは、モルヒネ鎮痛耐性形成の分子基盤の解明とその際の可塑的変調の可視化を試みてきた。モルヒネ鎮痛耐性の分子機構としてはモルヒネ適用時にアンチオピオイド機能の亢進が起因することを、遺伝子欠損動物を用いた解析から明らかにしてきていたが、さらに、この遺伝子欠損動物にその遺伝子を脳部位特異的にレスキューさせ、部位特異的な機能回復を確認するという新たな手法により、耐性形成に関わるアンチオピオイドシステムの役割を明らかにした。一方、慢性適用時に認められる可塑的神経回路変調の可視化においては、モルヒネ受容体のプロモーターで制御した小麦胚芽レクチン(WGA)と蛍光蛋白質の融合蛋白質遺伝子を脳の耐性形成や依存性形成に関わる領域に発現させ、シナプスを介する神経ネットワークを可視化することに成功した。また、モルヒネ慢性適用に伴うこの神経ネットワークの可塑的変調を可視化することにも成功し、現在論文作成中である。
