細胞分裂制御キナーゼの分子連携とその調節異常による腫瘍形成機構

研究概要	Auroraキナーゼは進化上高度に保存されている普遍的な分裂期キナーゼである。本研究は、詳細が不明であったAurora-Aの生理機能について調べ、ヒトの細胞分裂のメカニズムに迫ることを主題とした。本年度は、昨年度までに同定したAurora-Aの会合分子について、引き続き、それらの相互作用の解析を行った。さらに、最終年度として、これまでの結果を論文発表することも目標とした。以下に、本年度の成果を要約する。 (1)Aurora-Aは分裂期進入のための必須子である: リン酸化部位認識特異抗体を用いることにより、Aurora-Aの活性化がG2後期の中心体において始まることを見出した。そこで、RNAi法を用いてAurora-Aの発現を抑制したところ、HeLa細胞はG2期で停止し分裂期への進入が著しく損なわれた。細胞分裂の開始に際しCyclinB1-Cdk1の活性化が中心的な役割を持つと考えられ、その最初の活性化は中心体で起こるが、我々はG2期後期においてAurora-Aが中心体で活性化することが、つづくCyclinB1-Cdk1の活性化に必要であることを示した。 (2)AjubaはAurora-Aの自己リン酸化による酵素活性化を誘導する: Aurora-Aの会合分子としてヒトAjubaを同定した(GenBank Acc.No.AY169959)。Ajubaは3つのLIMドメインをもつタンパク質であり、LIM2およびLIM3を介してAurora-Aと結合することを見出した。分裂期においてAurora-AはAjubaと結合することで自己リン酸化を引き起こし、活性化することが明らかになった。AjubaはG2後期の中心体におけるAurora-Aの活性化を誘導する必須の調節因子であることが分かった。
