神経細胞におけるIP_3受容体の分子機構と情報伝達,発生・分化におけるその役割

研究概要	イノシト-ル3リン酸レセプタ-(IP_3ーR)は細胞内膜系、特に滑面小胞体に豊富に存在するCa^<++>チャネルである。まずIP_3ーRの構造・機能相関を解析した。IP_3ーR cDNAの欠失変異を作製して、NG108ー15細胞へ強制発現させて、IP_3との結合能を調べたところ、N末端650アミノ酸からなる配列内にあることが明らかとなった。更に膜貫通部位と推定されるC末端側の領域を欠失させると、膜へ組みこまれる可溶性蛋白質として回収された。しかもこの状態では単量体を形成し、IP_3結合能を有しているため、単量体でもIP_3結合能を有することが証明された。更に、膜貫通部位を欠失していても、そのC末端側を含んでいると、2量体を形成するので、IP_3ーRは、4量体を形成するために膜貫通部位とそのC末端側が重要であることが考えられた。次に外界の多様な刺激に対応してIP_3ーRに多様性があるか否かにつき検討をPCR方により加えた結果RNAのスプライシングによる数種のサブタイプがあることを明らかにした。スプライシングはN末端650アミノ酸のIP_3結合部位内(SI)と、Ca^<++>チャネルの調節領域と考えられる二つのリン酸化部位の間(SII)の2ケ書に存在した。IP_3ーRのうちでSIサブタイプはプルキンエ細胞がシナプス形成する時期にピ-クを示した。又SIIサブタイプを欠損したものは、変化しないが、SIIを含むものは、徐々に増加した。SIIサブタイプを含むものは中枢神経系のみにみられSIIは中枢神経、末梢組織を問わず普遍的に存在していたが、末梢組織はSIIを欠損したIP_3ーRのみを発現していた。この様にIP_3ーRは様々なアイソフォ-ムからなることが明らかとなり、4量体であることを考えると外界の多様性に対応して、IP_3ーRも多様性を示すことが明らかとなった。
