研究概要	IP3レセプターは細胞外からの刺激により産生されたセカンドメッセンジャーであるIP3と特異的に結合し、細胞内のCa^<2+>放出をおこなうリガンド依存性Ca^<2+>チャネルである。本年度はIP3レセプター解析をすすめ、以下の結果を得た。(1)我々が開発した三種類のモノクローナル抗体のうち、C末端側を認識する18A10抗体は、Ca^<2+>放出能のみを阻害することをミクロソーム画分を用いて明らかにした。更にこの抗体をハムスター卵に注入したところ、受精に伴う卵の活性化に重要なCa^<2+>波やCa^<2+>振動の現象が阻害された。更に、抗体存在下ではCa^<2+>やIP3をピペットで注入しても両反応が阻止されたことから、受精現象にIP3及び、IP3レセプターが関与することが明らかにされた。(2)ショージョーバエのIP3レセプターのcDNAをクローニングして、全塩基配列を決定した。マウスIP3レセプター(タイプ1)とアミノ酸で57%のホモロジーをもち、視覚、嗅覚などの感覚器官や筋組織に存在することが明らかになった。しかも、IP3レセプターのCa^<2+>放出の調節に重要と考えられる2ヶ所のセリン残基はショージョーバエのIP3レセプターにはなく、また両残基の中央に存在するスプライシングによる120塩基対はなく、マウスにおける末梢型タイプに属していると考えられた。(3)SH-SY5y細胞をムスカリン受容体のアゴニストであるカルバコール刺激して長時間PI代謝を亢進させると、IP3遊導Ca^<2+>放出能が低下したが、この現象がIP3レセプターそのものの減少によることを明らかにした。
