糖タンパク質糖鎖に含まれる情報の解析とその生理活性発現のメカニズムの研究

研究概要

1.hCGの糖鎖の機能
MAー10細胞を用いた健常妊娠,破壊奇胎患者,絨毛癌患者の尿から精製したhCGのホルモン活性の比較研究により、シアル酸以外の糖鎖構造の修飾の反合と平行してホルモン活性が低下することが判明した。
2.エリスロポエチン(EPOと略)糖鎖の機能
各種グリコシダ-ゼを用いたEPOの糖鎖修飾の研究により以下の点が明らかになった。1)ムチン型糖鎖の除去はEPOの<in>___ー <vitro>___ー活性に影響を与えないが,Asn型糖鎖の除去は活性の消失をひき起こす。2)Asn型糖鎖の側鎖に含まれるNeu5Ac,Gal,GlcNAcを順次除くと,EPOの<in>___ー <vitro>___ー活性は未処理に比べてそれぞれ3倍,4倍,5倍と上昇する。更にManαー,Manβーを除去すると活性は逆に減少する。3)上記のいかなるAsn型糖鎖の修飾も<in>___ー <vi>___ー <vo>___ー活性の消失をひき起こした。從ってマンノ-ス母核の先にNeu5Ac→Gal→GlcNAcという側鎖がついていることが,<in>___ー <vivo>___ーの活性発現に必須であり,マンノ-ス母核の存在が<in>___ー <vitro>___ーの活性発現に必要であることが確定したといえる。
3.リンホカインの活性発現における糖鎖の役割り
リンホカインの働きを阻害することで知られるウロモジュリンの糖鎖構造を調べ,GalNAcβ1→4Galを側鎖に持つ酸性の3本鎖,4本鎖が含まれることを明らかにした。次に破傷風毒素によるT細胞活性化をウロモジュリンが阻害するという知見をもとに、ウロモジュリンから遊離した糖鎖の作用を調べて,この阻害が糖鎖によるものであることを確認した。現在この阻害活性がどのような構造の糖鎖でひき起こされるかを調べている。
4.以上の研究に加えて本年度は白血球接着分子の糖鎖構造と機能の問題、受精初期反応における卵透明体糖タンパク質糖鎖の機能的役割の解明にも着手し研究を進めている。
