研究概要	長年捜し求められていたミオシン分子のATPによる構造変化をシンクロトロン放射X線小角散乱により、ついに捉えることに成功した:ミオシン分子のATPによる構造変化は、筋収縮の分子的過程の最も重要なものであり、多くのグループにより追求されてきたが、この変化を捉える事は出来なかった。我々は若林克三博士のグループと協力し、高速液クロにより精製した新鮮で純粋なミオシン頭部を用い、シンクロトロン放射X線小角散乱法により、ATPによって構造変化が生じることを突き止めることに成功した。部位特異的重原子標識電子顕微鏡法のためにディクチオ型粘菌のミオシン遺伝子に部位特異的変異を導入しディクチオ型粘菌で発現させたが、収量に問題があった。発現量を増す目的で、重鎖のC端部分を削り取りとった結果、数mgの変異ミオシンを精製できた。このミオシンはATP分解活性やアクチン結合能は保たれており、アクチンとの複合体の三次元再構成も可能であった。時間分解能電子顕微鏡法の開発が完成して、ケージドATPからのATP放出後、5ミリ秒から200ミリ秒のクライオ電子顕微鏡像を得ることに成功した。この時間分解電子顕微鏡像シリーズを解析して、ATPによるミオシンのアクチン・フィラメントからの離脱速度を単頭であるミオシンS1と双頭のHMMとで比較し、後者がより遅いことを確かめた。蛋白の電子顕微鏡法では、低電子線量での低雑音画像記録が重要である。イメージング・プレートを用いてトロポミオシン準結晶の電子顕微鏡像を1000電子/nm^2で記録し、非子午線情報も取り込んで雑音濾過し、トロポミオシンの一分子像を得ることに初めて成功した。トロポミオシン分子の電荷を持った部位のみを重原子染色したところ、X線結晶解析からの予測と良く一致する結果を得た。
