研究概要

1.Progression研究の実験モデル:同系宿主での造腫瘍性の極端に低下した癌細胞regressorを用いて、癌細胞の悪性化進展(progression)を検出する実験モデルを樹立した。すなわち、マウス線維肉腫由来QRからprogressionしたQRpPs、およびラット乳癌由来のERからprogressionしたERpPsはin vivoの造腫瘍性が増強するとともに浸潤・転移能が亢進した悪性化の進展した形質を獲得する。
2.Progressionの要因:(1)progression促進因子QR、ERのprogressionに宿主反応細胞が関与していることを明らかにした。QR、ERを宿主反応細胞と強制的に接触させると造腫瘍性が増強され、増殖してきた腫瘍にはprogressionしたクロ-ンが高頻度に出現する。QRのprogressionには急性期反応細胞の産出するフリ-ラジカルが、ERのそれには慢性期反応細胞の産出する増殖因子(EGF)がそれぞれ主として関与していることが示唆された。(2)progressionの癌細胞側の要因 癌細胞の遺伝的不安定性genetical instabilityがprogressionしやすい原因と考えられているが、癌細胞の遺伝的不安定性を解析する実験モデルが得られている。すなわち、QRのうち、容易にprogressionするQRー32とprogressionしがたいQRー29があることが注目された。
3.Progressionした癌細胞の性格:progressionしたQRpP、ERpPは浸潤・転移能が亢進しているほか、(1)細胞膜面のmicrovilliの密な形成、(2)細胞骨格を形成するアクチンとそれに関連した蛋白の異常、(3)免疫抑制因子であるPGE_2の大量産生、(4)ガングリオシドGM3のsheddingの亢進、(5)増殖因子受容体の遺伝子、蛋白レベルでの発現の増強とともに、(6)染色体およびDNA ploidy patternの変化が認められ、遺伝子のレベルでの変化も示唆されている。このことは、本研究で人為的にprogressionさせた癌細胞が、臨床的にprogressionした癌細胞と同様な遺伝子レベルでの変化を伴なった諸変化をおこしていることを示唆する。
