放射線抵抗性癌の成因とその増感

研究概要	本研究の目的は、放射線抵抗性癌の原因が、癌細胞の高い修復能力によるものか、高い防御機構によるものか、また活性化癌遺伝子の発現によるものか、その癌遺伝子の役割は何か等を明らかにすることによって、その増感方法を見い出し、より戦略的な放射線治療の基礎資料を提供することにある。研究方法:動物やヒトの抵抗性癌を用いて検討する。その方法は、in vitroのコロニ-形成法、in vivoのモデル系としてのスフェロイド法等で検討する。また亜致死障害(SLD)や潜在致死障害(PLD)修復をさせた場合に、放射線抵抗性になるのかどうかも検討する。その抵抗性の機構としては、活性化rasやraf癌遺伝子の発現の有無、DNAレベルの修復能、細胞内のスカベンジャ-量等について検討する。研究成果:ヒト骨肉腫細胞株(9株)は、ほとんどが抵抗性であったため、DoとDqから見て、特に感受性には大きな差が見られなかった。しかし、PLD修復能の非常に高い(生存率が15倍ほど上昇する)細胞株もあり、この高いPLD修復能ゆえにヒト骨肉腫が放射線抵抗性になっている可能性が示唆された。骨肉腫細胞が正常線維芽細胞に比べSLD修復能が高いことから、治療において2Gyを分割照射において抵抗性になる原因なのかも知れない。ヒト骨肉腫細胞においてその放射線抵抗性とras癌遺伝子との関連について検討したが、少なくとも調べたヒト骨肉腫ではras遺伝子の活性化と放射線抵抗性とは、対応していないと結論された。一方、細胞内のGSHが放射線感受性と関係していることを示唆する結果を得た。ゴ-ルデンハムスタ-由来のファイブロサルコ-マ細胞株(5株)において、放射線感受性の指標値であるDo値と初期DNA二重鎖切断量とが良い相関を示したが、修復不能DNA二重鎖切断量との相関は見られなかった。
