研究概要	活性化癌遺伝子を導入すると大体予測された細胞の癌化がおこる。しかし、活性化Hーrasでは胚性腫瘍が、Nーmycでは発生異常がおこる。また、フレンドウイルスのgp55をβーアクチンプロモ-タ-を用いて種々の組織で発現させても、赤芽球腫はおこるが他の細胞の癌化はおこらない。このことは、細胞癌化のためには、もともとその細胞において情報伝達に関与しているセットの遺伝子の活性化が必要なことを示唆している。活性化癌遺伝子による腫瘍化では殆んど単クロ-ン性であり、他の第2次、第3次の変化が生じる必要のあることが示唆されている。アルブミンプロモ-タ-を接続したSV40T抗原遺伝子を導入すると肝癌が生じるが、この癌細胞ではHーrasの変異もおこっていることが確認された。また、Eμーmyc遺伝子によるリンパ腫の場合、βリンパ腫では第4番染色体の異常が、C3HマウスにおけるTリンパ腫の場合は第6番染色体の異常が生じていることが明らかとなった。TL抗原遺伝子の導入では、γδT細胞のリンパ腫が高頻度に生じるが、これらの癌細胞では20%に新たなレトロウイルスの組込みが観察された。これらの変化が2次要因として癌化に関与している可能性が示唆された。癌遺伝子や癌抑制遺伝子以外の要因も癌の発生に関与している可能性がある。これらの要因の中でDNA修復に関与するメチル基転移酵素の影響を調べるため、大腸菌の遺伝子を導入したマウスを作製したところ、自然発生の肝癌の頻度が低下している可能性が示唆された。また、βーインタ-フェロンの強制発現によっても肝癌の頻度は低下していた。更に、Eμーmycによるリンパ腫の発生において、用いるマウスの遺伝背景によって、発症年令や癌化する細胞型が影響をうけることも明らかとなった。野性マウスを用いた研究から、染色体の組換えを高頻度におこすDNA領域が存在することが明らかとなった。ES細胞を用いた相同遺伝子組換えにも成功した。
