研究概要

本年度は今後の研究を遂行するために必要な実験材料や実験系の準備に重点をおいて各班員が研究を行った。この結果、免疫グロブリン遺伝子、CD2遺伝子、クラスII遺伝子を導入したトランスジェニックマウスが今後の研究に利用できるようになった。同時に、T細胞レセプタ-遺伝子を導入したトランスジェニックマウスも現在準備中であるが、本年度には完成できなかった。次に、骨髄内でのリンパ球や血液幹細胞の増殖に関わる分子に対するモノクロ-ナル抗体作成を試み、抗cーkit抗体、抗ストロ-マ細胞抗体が確立され、今後の研究の進展に大きく貢献することが期待される。一方、胸腺内T細胞の増殖分化の研究は少し進展が遅れている。しかし、先述のT細胞レセプタ-遺伝子導入トランスジェニックマウスの準備は着々と進んでいるし、T細胞分化増殖を支持できるストロ-マ細胞株についても2株本年度に準備できた。また、T細胞とストロ-マ細胞との相互作用に関与していることが予想される様々な細胞接着分子に対するモノクロ-ナル抗体も既に作成され、今後これらを用いた研究が進むと期待される。最後に、リンパ球への分化能を有する幹細胞株を用いた実験系も確立でき、現在この系でみられる分化の各ステップに発現している遺伝子について研究が協同で進められるようになっている。
この様に、材料や実験系の開発も含めて進めていく事が求められる長期計画の最初の段階は満足いく進展が得られたと考えている。また、骨髄内での細胞増殖については予想以上の成果が得られた。このため、来年度では最初全く計画していなかった、幹細胞からの分化決定の分子機構についても計画に取り込んで研究を進めて行きたいと考えている。
