非免疫系抗腫瘍性因子の構造と機能

研究概要

本年度は、各非免疫系抗腫瘍性因子を精製し、物理化学的性状を明らかにすることを目標においた。それぞれの因子によって、進展の度合いに多少の差異はあるが、ほぼ目標の達成ができた。それぞれの因子について以下に述べてみる。腫瘍変性因子については、ヒト線維芽細胞の培養上清を、Sーセファロ-ス、Con Aーセファロ-ス、ヘパリンカラムクロマトグラフィ-にかけてみたところ、ほぼ単一バンドが得られるようになった。この物質のアミノ酸配列を現在解析しているところである。軟骨由来抗腫瘍性因子については、コラゲナ-ゼインヒビタ-であるTIMPおよびTIMPー2と比較検討することにより、物理化学的性状を解析した。その結果、抗腫瘍性因子の1部は、TIMPではないかと推察された。ウサギ血清中の増殖阻害因子については、各種イオン交換カラムクロマトグラフィ-、アフィニティ-クロマトグラフィ-などを用いて10万倍精製度を上げることができ、単一のバンドになった。新生期脳由来抗癌因子については、すでに単一バンドとして精製されているが、今回はこれに対する抗体を作成し、生理的役割を解析した。グリア細胞成長因子については、Schwannoma細胞に対し分化誘導することが確認された。また、神経線維腫から抽出されたneurotrophic factor(NTF)が、神経芽細胞腫のDNA合成を促進すること、同じ抽出物中にグリア細胞増殖抑制因子が存在することが明らかにされ、精製を行った。腫瘍退縮因子については、作用機作として、好中球の遊走を増強することがわかり、さらに腫瘍退縮因子の拮抗因子の精製も行った。センチニクバエの抗腫瘍活性を持った因子は、分子量が200kであることがわかり、単一バンドにまで精製できた。そのほか、線維芽細胞から分泌される分化誘導因子を単一バンドまで精製を行い、アミノ酸配列を解析したところ、IL6に一致することがわかった。
