膵癌発生の機構解析とその制御に関する研究

研究概要	分担研究者は、各自の課題のもとに以下の成果を得た。小西は、ハムスタ-を用いてNーnitrosobis(2ーhydroxypropyl)amine(BHP)により膵発癌をイニシエイトした後にCCKの拮抗薬であるproglumide(PGL)を投与してその膵発癌に対する修飾効果を検索したが、PGLによる修飾効果は見出せなかった。又、ハムスタ-にNーnitrosobis(2ーoxopropyl)amine(BOP)にて膵発癌をイニシエイトした後に天然抗酸化剤であるカテコ-ル、ハイドロキノン、レゾルシノ-ルを食餌性に投与すると、0.75%と1.5%のカテコ-ル、1.5%ハイドロキノンとレゾルシノ-ルとはハムスタ-1個体当りの膵病変の発生を抑制した。高橋は、ハムスタ-にBOPを投与して膵発癌をイニシエイトすると同時にトリプシン阻害剤であるカモスタットを50ppmの用量で投与すると膵発癌の抑制されることを見出した。関谷は、PCRーSSCP法によりハムスタ-可移植系膵癌においてcーKーrasのexon1における遺伝子異常を検出した。更に、BHPによるハムスタ-原発膵癌にて14匹中14匹にcーKーrasのexon1における遺伝子異常の存在を明らかにした。森は、(1ー^<14>C)ーBHPのハムスタ-における臓器特異性代謝産物を検索した結果、Nーnitroso(2ーhydroxypropylーoxopyl)amine(HPOP)が膵を含む各臓器における主要代謝産物であることを明らかにした。鎌野は、イヌにおけるENNGによる膵管腺癌発生系において内視鏡的逆行性胆膵管造影にて膵管の変化を詳細に観察して病理組織学と対立することにより膵管上皮細胞の過形成が前癌病変として存在することを見出した。石川は、膵癌患者では血中プロインスリンが高値で、インスリンのCーペプタイトが低値であることを見出し、その比を算出することにより早期膵癌の診断に対する有用性を示した。澤武は、血液型関連糖鎖抗原II型に属するシアリイル・ルイスメは膵癌患者に特異的且つ高率に出現することを示した。
