研究概要	本研究班の平成2年度の研究概要を以下に記す。分担研究者名を( )内に記す。 1.マウス皮膚2段階発癌モデルを用いて、パピロ-マ退縮後の正常化した皮膚におけるHーras遺伝子の点突然変異を検出した(小川)。 2.酸化還元酵素系修飾下に内因性に産生される活性酵素により誘発されるDNA損傷についてラット肝を用いて調べた(傳田)。 3.膀胱発癌系でDMABーDNA付加体の形成は酸化防止剤の併用投与群で約2倍に増加した(白井)。 4.発癌物質4HAQOを4種類の動物に投与し、DNA付加体とDNA修復を計測して定量し、発癌の種、臓器特異性との相関を明らかにした(石川)。 5.O^6ーアルキルグアニンの超高感度定量法を考案し、数fmolレベルで定量することが可能となった(許)。 6.DNAの高次構造の変化をCDスペクトルや抗ZーDNA抗体を応用して、AAFや4NQOがDNAの修復に及ぼす変化を検出(多田)。 7.C3Hマウス由来の肝細胞培養系を樹立し、活性化rasをトランスフェクトすると可移植性になることがわかった(北川)。 8.cーmycの発現を負に制御する領域について、モリス肝癌のエクソン3に1塩基挿入の突然変異を見いだした(林)。 9.メチルトランスフェラ-ゼ完全欠損株で、プラズミドに生じた変異をしらべるとGC→ATの塩基置換がホットスポットに集中してみとめられた(真木)。 10.G_1期の進行に欠損をもつハムスタ-の変異性を相補するヒトの遺伝子のクロ-ニングに着手した(山下)。
