研究概要	基礎的研究:真核DNAトポイソメラ-ゼ(トポ)IのN端側の機能領域が狭められた。リン酸化によるトポIIの活性調節では相矛盾する結果を与え、結論に至っていない。ラットにおいてトポIIAとトポIIBの2種類のアイソザイムがある事が示された。DNAのトポロジ-と機能については、核マトリックス上にSAR領域を介してDNAを結合する120Kのタンパクがあり、cDNAが分離された。また超ラセンDNAを特異的にマトリックスに結合させる75Kタンパクが精製された。そして構造が解析されつつある。またDNAに超ラセンを導入し転写効率を上昇させるタンパク因子が精製された。応用的研究:抗癌剤MSTー16及び関連化合物が既知のトポII標的抗癌剤とは異なる新たな作用機作を持つトポII阻害剤である事が発見された。細胞に対してはM期特異的阻害を示した。これは哺乳類においてもトポIIの必須機能はM期における染色体の凝縮、伸展、分離の各段階にある事を強く示唆している。トポ標的抗癌剤に対する耐性獲得機構に関しては以下のようである。カンプトテシン耐性には二つの型があった。第一はトポI遺伝子変異による酵素の薬剤耐性化、第二は酵素の発現レベルの低下によるものであった。トポII標的薬剤(VPー16)では酵素の発現レベルの低下による耐性獲得であった。さて、抗癌剤の臨床使用に当たっては併用療法が通常である。上記基礎的及び応用的研究をふまえて、トポ標的薬剤を中心としたベストの組合せ、投与法を案出すべく、in vitro培養で検討した。その結果、細菌のトポIIであるジャイレ-スの阻害剤ニュ-キノロン系薬剤がカンプトテシンー11の殺細胞効果を濃度依存的、時間依存的に増強した。以上、基礎、応用両面において本年度は著しい進展が見られた。
