研究概要	1.rasと関連のあるタンパク質であるGAPとホスホリパ-ゼCの大腸腫瘍における異常大腸がんではKーras遺伝子の点突然変異の頻度が高いので、ras周辺のタンパク質の異常を調べた。GAP(GTP aseーactivating protein)は家族性大腸腺腫症(FAP)由来を含む大腸がんの細胞のいくつかで活性が高かった。GAPの活性の高い細胞ではKーrasの点突然変異がなく、逆にGAP活性が検出されなかった細胞ではKーrasに変異があった。ホスホリパ-ゼC活性は大腸がん細胞のいくつかで高かった。特にFAP由来大腸がん細胞では正常の3〜4倍の上昇が認められた。 2.ヌ-ドマウスを使う方法によりFAP患者細胞DNAから得られたがん遺伝子の解析このがん遺伝子のcDNAクロ-ニングに成功した。一部、DNA塩基配列を決定してホモロジ-検索を行ったところ、Bーraf遺伝子と同一であることがわかった。cDNAの5'側はBーraf protoーoncogeneの配列とは異なっていたので、再配列が起こっていると考えられる。 3.大腸がんの大部分は腺腫由来かどうかの検討大腸がんの発生母地として、腺腫と正常粘膜のどちらが多いかを遺伝子レベルで解析するため、腺腫・がんについてKーrasの点突然変異の頻度を調べた。その結果、Kーras遺伝子コドン12の点突然変異はFAP症例と非遺伝性症例とを合計すると、正常粘膜と軽度異型腺腫では0%、中等度異型腺腫では8.1%、高度異型腺腫では83.3%、進行がんでは26.0%に発生しており、進行がんでは高度異型腺腫に比して変異率が有意に低かった。大腸がんの大多数が高度異型腺腫から発生するとするなら、進行がんのKーrasの変異発生率は高度異型腺腫と同程度か、更に高いはずである。故に腺腫からの発がんは大腸がん発症の主経路ではないと考えられる。
