研究概要	我々はRBタンパク質がM期特異的cdc2キナ-ゼによってin vitroでリン酸化されることを以前に示してきたが,in vivoではRBタンパク質のリン酸化はG_1/S期の境界線から始まりM期まで続く。そして,RBタンパク質による増殖抑制活性がこのリン酸化で解除されると推定される。そこで,G_1/S期特異的なものを含むすべてのRBキナ-ゼを検出することを試みた。その過程で,マウスFM3A細胞のcdc2キナ-ゼがMonoSカラムクロマトグラフィ-でG_1/SとG_2/M特異的な2つのピ-クに分かれ,RBキナ-ゼのピ-クはその2つの間に現れることを見出した。RBタンパク質上にはcdc2キナ-ゼの基質になるための共通配列をもつ部位が11ケ所に存在するが,それらの部位を含む合成ペプチドはすべてこのRBキナ-ゼのピ-ク分画でリン酸化された。さらに,このピ-クとほぼ重なる領域にサイクリンAが検出された。一方,RBタンパク質をリン酸化するキナ-ゼが抗RB抗体によってRBタンパク質と共沈殿されることも見出した。その活性は特にG_1/S期に強いこともわかった。以上の結果はG_1/S期にRBタンパク質をリン酸化する酵素がcdc2キナ-ゼであることを示唆しているけれども,in vivoとin vitroでリン酸化部位が一致するか否か,さらに,そのcdc2キナ-ゼに含まれているサイクリンの種類なども今後明らかにせねばならない。また,我々は,RBタンパク質のC末端から317個のアミノ酸をもつポリペプチドを大腸菌で発現させ,これを用いてRBタンパク質に対するモノクロ-ナル抗体を作製することにも成功した。これらの抗体はRBタンパク質の免疫沈降にも使えることがわかった。
