研究概要	リグノセルロ-スからの微生物によるエタノ-ル生産プロセスを開発するため、リグノセルロ-スの前処理と生成セルロ-スとキシランを直接エタノ-ル発酵する菌の育種について研究を行い、以下の結果を得た。リグノセルロ-スの前処理として蒸煮・爆砕処理と過酸化水素・アルカリ処理を併用した複合処理の効果を検討し、蒸煮・爆砕単独処理の場合に比べて2〜3倍の高い酵素糖化率を得た(前川英一)。爆砕稲わらの抽出成分量、細孔径分布、酵素糖化率やアルコ-ル転換率を検討し、爆砕稲わらの糖化発酵には蒸気圧3.3〜3.8MPa蒸煮時間1.0〜3.5minの爆砕が最も効果的であるという結果を得た(沢田達郎)。担子菌Bjerkandera adustaのcDNAおよび染色体DNAからリグニンペルオキシダ-ゼ遺伝子をクロ-ニングし、その塩基配列を決定した。また培養にポリウレタンフォ-ムを用いて、本酵素の生産性を向上させた(桑原正章)。 Saccharomyces cerevisiaeにクロ-ニングするためのセルラ-ゼ遺伝子の供与体としてセルロ-ス資化性の高い酵母を分離し、精製酵素遺伝子のクロ-ニングを行い数個のポジティブクロ-ンを得た(宮川都吉)。枯草菌由来の中性セルラ-ゼと好アルカリ菌由来のアルカリセルラ-ゼを材料に用い、蛋白工学的手法により、pH4〜5の酸性領域での酵素活性に寄与する5個のアミノ酸残基を同定した(堀之内末治)。 Bacillus pumilisのβーキシロシダ-ゼ遺伝子(xynB)を大腸菌の中で高発現させ、その遺伝子産物を単一にまで精製し、性質を検討したところキシロビオ-スに対して既知の酵素と比較して高い活性を示すことを認めた(卜部格)。キシロ-スリダクタ-ゼとキシリト-ルデヒドロゲナ-ゼの遺伝子をS.cerevisiaeにクロ-ニングし高い酵素活性が得られたが、キシロ-スからエタノ-ルを生産したりキシロ-スを炭素源として増殖するには至らなかった(吉田敏臣)。
