研究概要

本研究では、生体において免疫担当細胞等が活性化される機構を分子レベルで理解し、それに基づいて材料化学の立場から、新しい生体および細胞賦活材料の作製を推進することを目的としている。
1.PEGー脂質を組み込んだリポソ-ムが、融合機能を有することを見出し、植物プロトプラストとも効率よく融合し得ることを実証、細胞工学での新しい基礎技術を提供した。(砂本)
2.非ステロイド系抗炎症薬および局所麻酔薬等に対するテルペン系化合物の経皮吸収促進作用をスクリ-ニングした結果、炭化水素系の単環モノテルペン類に顕著な活性を見い出した。活性の発現には、水・エタノ-ル混合溶液の組成比が大きく影響し、このような溶液を大量に保持できる粘着性高分子素材としてアクリル酸素のポリマ-が適当であることを確認した。(永井)
3.温度変化に対して大きな膨潤変化を示すNーイソプロピルアクリルアミド(IPAAm)とアルキルメタクリレ-ト(RAA)のコポリマ-ゲルを用いて、薬物放出デバイスを作製し、外部温度変化による薬物放出のonーoff制御機能を検討した。本年度は特に、ポリマ-のアルキル鎖長を系統的に変え、onーoff放出における温度応答性を詳細に検討した。その結果、アルキル鎖長変化は、薬物放出速度および表面収縮層の生成過種に影響を与え、パルス型薬物放出パタ-ンを制御するうえで重要な因子となることを明らかにした。(岡野)
4.MDPのDーグルコ-スアナログ(GADP)をカ-ドランおよびキチンのオリゴマ-にコンジュゲ-ト化した新しいタイプのハイブリッド型免疫賦活抗腫瘍剤の合成法を確立した。(大内)
