研究概要	薬物・核酸・タンパク質を封入したリポソ-ム表面に、弱酸性pH(<6.0)領域でのみ膜融合活性を持つペプチドを結合させた人工ウイルスを調製し、細胞のエンドサイト-シスを利用した効率の良い、細胞にダメ-ジの少ないドラッグデリバリ-システムの開発を目的とし、本年度は次の(1)(2)の結果を得た。 (1)人工ウイルス表面のfusogenとして最も重要な弱酸性pHで正確に膜融合活性が制御できるペプチドを、インフルエンザウイルスの膜融合タンパク質(HAタンパク質)の活性部位を模したHAーペプチドをプロトタイプとして5種合成した。これらペプチドによる中性リン脂質(egg PC)よりなるリポソ-ムの融合活性を測定し、また、ペプチドとリポソ-ム・赤血球膜などとの相互作用を円二色性偏光など物理化学的手法を用いて研究した。その結果、各ペプチドは至適pHを5.0付近に持ち、pH6.0以下でのみ効率の良い膜融合活性・赤血球溶血活性などを持つことが判った。また、活性の無い中性pHにおいても強くリポソ-ムの膜表面に結合していること、酸性でのペプチド中の酸性アミノ酸残基のプロトン化と、それによるペプチドの疎水性の増加がペプチドのヘリックス含量を増加させ、活性発現の引金となることなども判った。これらのことより、今後、fusogenとなるペプチドは「両親媒性ヘリカル構造」を基本構造とする酸性ペプチドとして設計すればよいことが示された。 (2)人工ウイルスの生きた培養細胞内での挙動を経時的に追跡する手法の確立は、設計された人工ウイルスの検定に必要不可欠である。我々は、セルソ-タ、時間分解顕微蛍光法などを利用し、培養CHO細胞にエンドサイト-シスさせた蛍光性リポソ-ムを用いて、蛍光の共鳴エネルギ-移動効率の変化により、そのリポソ-ムの細胞内エンドソ-ム、リポソ-ム内での分解・融合の程度を顕微鏡下に生きた細胞を観察しながら定量的に測定する方法を開発した。
