研究概要

本研究は、計画研究6班の総括班として、以下の活動を行った。1.総括班会議は平成2年8月と平成3年1月の2回東京で開催し、運営に関する諸問題を審議した。2.各計画研究班の最も著名な成果を中心に平成3年1月10日に東京でシンポジウムを開催し、約150名の参加を得て好評であった。3.本重点領域研究の包含する広範な分野を相互に紹介し理解と交流を深める目的で、班員の和文総説の別刷を選んで集録し、複写製本して班員に配布した。4.計画研究6班の班間にまたがる交流を促進するため、班間共同研究を公募し、非常に有意義と認められた5件を財政的に支援した。5.総括班員の創意により、長期的展望の下に、各計画研究班では実行の困難ないくつかの試行錯誤的研究を行った。以下に二、三の著るしい成果を挙げる。(1)橘は、細胞内情報伝達過程において従来ほとんど注目されていなかったMgイオンに注目し、蛍光色素magーfuraー2と蛍光顕微画像解析装置を用いて単一細胞内遊離Mgイオン濃度の測定法の確立に成功し、それを用いてマウスSwiss 3T3細胞をボンベシン+インスリン等で増殖刺激すると、30分から1時間以内に細胞内遊離Mgイオン濃度が上昇することを認めた。したがって細胞内遊離Mgイオン濃度の変化が修飾因子として細胞活動を規定している可能性が考えられ、今後のこの方向の発展が期待される。(2)三品はこれまで多くの研究者が試みて来たが誰も未だ成功していないグルタミン酸受容体の二種類のサブユニットのcDNAクロ-ニングとその発現に成功し、その機能を解析して、カイニン酸とキスカル酸の受容体は同一分子であること、グルタミン酸受容体は複数のサブユニットから構成されていることなど、非常に興味ある知見を明らかにしつつある。
